
Plasma Extraction
Plasma: This standard procedure calls for extraction with acetonitrile (3 times 3:1 acetonitrile:sample). The acetonitrile crashes out plasma proteins after the first addition, giving a very gummy protein pellet that is difficult to break-up for further extractions.
Current protocol 3:1 acetonitrile, vortex spatula.
By using Covaris acoustics, it is possible to greatly improve upon the traditional methods of plasma extraction. By reducing the ratio of Acetonitrile/Plasma to 1:1, then acoustically mixing, this creates a very homogenous flocculent prep. Then 2 parts of acetonitrile are added for the final extraction.
It is also possible to spin down the pellet, cryo-pulverize it to rapidly shatter the pellet, and then carry out extraction on the increased surface area.
By carrying out the extraction in two parts there is NO gummy material. The final constituency is very flocculent, with a large surface area allowing highly efficient repeatable extractions.
